The E7B6 peptide epitope is an 18-amino acid sequence (EKEALKKIIEDQQESLNK) derived from the human nuclear autoantigen La, also known as Sjögren's syndrome-associated antigen B (SS-B) [1, 6]. It corresponds to residues 311–328 of the native La protein and is characterized as a cryptic epitope, meaning it is hidden within the protein's native structure and only becomes accessible upon denaturation or when expressed as an isolated peptide [1, 9]. In the field of immunotherapy, E7B6 is primarily employed as a modular "tag" or "switch" within advanced chimeric antigen receptor (CAR) T-cell platforms, such as the UniCAR and RevCAR systems [2, 5]. In these applications, the epitope is expressed on the surface of engineered T cells or integrated into bispecific adapter molecules (Target Modules) to allow for the precise, dose-dependent control of T-cell activation against various tumor-associated antigens [4, 10]. Additionally, the E7B6 epitope can function as an "elimination tag," enabling the selective destruction of CAR T cells by anti-E7B6-directed effector cells to mitigate severe treatment-related toxicities like cytokine release syndrome [3, 6].
