Overview

La ribonucleoprotein 1, translational regulator (LARP1) is an RNA-binding protein that plays a key role in post-transcriptional gene regulation, especially as a downstream effector of the mTORC1 signaling pathway. It binds to the 3' poly-A tails of mRNAs and the 5'-cap of mRNAs containing a 5' terminal oligopyrimidine (5'TOP) motif—primarily genes encoding ribosomal proteins and factors involved in translation. LARP1 modulates translation by repressing or promoting ribosome recruitment to these mRNAs, thus influencing protein synthesis rates according to cellular growth signals and nutrient availability. It is expressed at low levels in most normal tissues but is highly upregulated in various cancers, and high expression correlates with poor prognosis. LARP1 also enhances the replication of dengue virus and may have roles in the cellular stress response.

Mechanism of action

Inhibition or modulation of mTORC1 signaling leads to altered LARP1 phosphorylation and changes translational regulation. Drugs that target upstream pathways (mTOR inhibitors) may reduce LARP1-mediated translation repression of TOP mRNAs.
