Overview

LacdiNAc (GalNAcβ1-4GlcNAc) is a terminal disaccharide epitope found on N-linked and O-linked glycoproteins as well as glycosphingolipids (Nyame et al., 2004). In normal human physiology, its expression is highly restricted, notably appearing on pituitary gonadotropins like luteinizing hormone (LH), where it regulates circulatory half-life through recognition by the hepatic GalNAc-4-SO4 receptor (Baenziger, 1994). However, LacdiNAc is significantly upregulated in several malignancies, including prostate, ovarian, and pancreatic cancers, where it is considered a tumor-associated carbohydrate antigen (TACA) linked to progression and metastasis (Miura et al., 2016). It also serves as a major immunodominant antigen in helminth parasites such as Schistosoma mansoni, facilitating host-parasite interactions and immune evasion (Smit et al., 2015). Therapeutic strategies targeting LacdiNAc include the use of specific lectins like Wisteria floribunda agglutinin (WFA) for diagnostic imaging and the development of monoclonal antibodies or glycan-based vaccines for cancer and parasitic infections (Merten et al., 2022).

Binding to the terminal GalNAcβ1-4GlcNAc moiety to block lectin-mediated cell signaling, facilitate diagnostic detection via lectin-affinity, or induce immune-mediated destruction of target cells via antibody-dependent mechanisms.
