Overview

Ladinin-1 (LAD1) is a collagenous anchoring filament protein primarily localized to basement membranes, where it plays a vital role in the integrity and cohesion of tissue boundaries, particularly at the dermal-epidermal junction[1][4][5][6]. It ensures stable adhesion between epithelial layers and the mesenchyme. LAD1 is dynamically regulated in response to various physiological and pathological signals, including glucocorticoid receptor signaling. Pathologically, LAD1 is notable as a major autoantigen in linear IgA bullous dermatosis, an autoimmune blistering skin disorder[1][4]. Additionally, its dysregulation is implicated in tumor proliferation, migration, and metastasis, particularly in epithelial cancers such as oral squamous cell carcinoma, prostate cancer, and laryngeal squamous cell carcinoma[1][2]. LAD1 is considered a potential prognostic biomarker and a putative therapeutic target due to its dual role as a structural protein and immunologically reactive molecule[1][2][4].
