Overview

Laminin-332 is a heterotrimeric glycoprotein essential for the structural integrity of the dermal-epidermal junction (UniProt P19137, Q13751, Q13753). It acts as a bridge, connecting basal keratinocytes via integrins to the underlying dermis through its interaction with Collagen type VII alpha 1 chain (COL7A1) (Rousselle et al., 1997). Specifically, the interaction between the alpha-3 chain of Laminin-332 and the von Willebrand factor A-like (vWFA) subdomains, particularly vWFA2, within the non-collagenous 1 (NC1) domain of Collagen VII is critical for the assembly of anchoring fibrils (Tufa et al., 2019). Defects in this interaction, caused by mutations in either the LAMA3, LAMB3, LAMC2, or COL7A1 genes, result in severe mechanobullous diseases like Epidermolysis Bullosa (PubMed: 29431077). Therapeutic interventions focus on restoring this protein-protein interaction through gene therapy (e.g., Beremagene geperpavec), recombinant protein replacement (e.g., PTR-01), or cell-based approaches to prevent skin fragility and chronic ulceration (FDA, 2023).
