Overview

Laminin-411, formerly known as Laminin-8, is a heterotrimeric extracellular matrix protein composed of the alpha-4 (LAMA4), beta-1 (LAMB1), and gamma-1 (LAMC1) chains (UniProt P50579, P07942). It is a primary component of the vascular basement membrane, particularly in the endothelial cells of the blood-brain barrier (BBB) (Sorokin, 2010). Laminin-411 plays a critical role in facilitating the migration of leukocytes, specifically Th17 cells, from the blood into the central nervous system by interacting with receptors such as integrin alpha-6 beta-1 (Wu et al., 2009). In diseases like multiple sclerosis, the expression of Laminin-411 is often upregulated, promoting neuroinflammation. It is also implicated in tumor angiogenesis and the progression of high-grade gliomas, where its expression correlates with tumor grade (Ljubimova et al., 2001). While there are currently no FDA-approved drugs targeting Laminin-411 or its encoding mRNAs, it remains a significant area of research for therapeutic interventions aimed at stabilizing the BBB or inhibiting pathological cell migration.
