Overview

Laminin protein refers to a family of large, heterotrimeric glycoproteins that are a fundamental component of the basement membrane in all animals[1][2][3][5][6]. Each laminin molecule is composed of three subunits (alpha, beta, and gamma chains) assembled together into a cross-shaped structure, enabling interactions with other extracellular matrix components and cell surface receptors such as integrins and dystroglycan[1][2][3][4][6]. In humans, 15 distinct laminin heterotrimers have been biochemically defined from combinations of 5 alpha, 4 beta, and 3 gamma chains[1][3]. Laminins are key for the assembly, structural integrity, and function of basement membranes, guiding cell adhesion, migration, differentiation, and tissue morphogenesis[1][2][3][5][7]. Mutations or dysfunction in specific laminin chains are associated with diseases such as congenital muscular dystrophy, Pierson syndrome, certain forms of epidermolysis bullosa, and can influence cancer progression[5][6]. Laminins are not classical therapeutic targets like receptors or enzymes, but their domains are of interest in regenerative medicine and as disease biomarkers[5][6].
