Overview

Laminin-111, historically referred to as Laminin-1, is a large heterotrimeric glycoprotein consisting of alpha-1, beta-1, and gamma-1 chains. It is one of the first extracellular matrix proteins expressed during embryogenesis and is essential for basement membrane assembly and early development (UniProt P12601, P07942, P11047). Biologically, it serves as a scaffold for cell attachment and influences diverse processes such as cell differentiation, migration, and neurite outgrowth through its interactions with integrins and dystroglycan (PubMed: 19433786). In the context of disease, Laminin-111 is a significant therapeutic target for Duchenne Muscular Dystrophy (DMD), as its exogenous administration can functionally substitute for missing laminin-211 to stabilize the sarcolemma (PNAS: 106(19)). It also plays a complex role in oncology, where it can either restrict or facilitate tumor cell invasion depending on the microenvironment (NIH: Laminin). Therapeutic strategies involving Laminin-111 primarily focus on protein replacement or the use of its active peptide fragments to promote tissue repair and regeneration.
