Overview

Laminin subunit alpha-4 (LAMA4) is one of the three components of laminin heterotrimers, key extracellular matrix glycoproteins that form the major noncollagenous constituent of basement membranes[1][2][3][5]. The protein supports cell adhesion, migration, differentiation, and signaling across multiple tissues and organ systems. In particular, LAMA4 interacts with integrins and integrin-linked kinase (ILK) to maintain endothelial integrity and myocardial performance. Deficiency or mutation disrupts cell-matrix interactions, resulting in loss of endothelial cells, cardiomyopathy, and abnormal angiogenesis in the retina[2]. The exact tissue-specific and isoform-specific functions of LAMA4 remain incompletely understood, but its structural and signaling roles in vessel morphogenesis and tissue homeostasis are well documented[2][3][5].
