Overview

Laminin subunit beta-3 (LAMB3) is a structural protein encoded by the LAMB3 gene, serving as the beta chain of laminin-332, a major component of the basement membrane underlying epithelial tissues such as skin. Laminin-332 (formerly laminin-5), composed of alpha-3, beta-3, and gamma-2 chains, is essential for cell adhesion, migration, and the formation of the anchoring filaments that stabilize the epidermal-dermal junction. LAMB3 plays critical roles in the maintenance and repair of epithelial barriers, wound healing, and organ morphogenesis. Mutations in LAMB3 cause junctional epidermolysis bullosa, a severe inherited disorder characterized by skin fragility and blister formation. Aberrant expression is linked to malignant progression and poor prognosis in several cancer types, and is being investigated as a prognostic biomarker and potential therapeutic target in oncology and inflammatory diseases[1][3][4][7][8][10].
