Overview

Laminin subunit gamma-3 (LAMC3) is a protein encoded by the human LAMC3 gene and represents the gamma-3 chain component of the heterotrimeric laminins, a family of extracellular matrix glycoproteins essential for basement membrane structure and function. Laminin gamma-3 is broadly expressed in tissues such as skin, heart, lung, and reproductive tracts, with notable abundance at the apical surface of ciliated epithelial cells in organs like the lung and reproductive system, where it contributes to the morphogenesis and structural integrity of ciliated processes. It is also a key component of laminin-12 and plays roles in cell adhesion, migration, differentiation, and structural support. LAMC3 is required for proper cortical development in the brain, and mutations in this gene are associated with cortical malformation diseases (polymicrogyria, pachygyria) and global developmental delays. Genetic associations with neurodegenerative disease have also been suggested, but LAMC3 is not known to be a direct therapeutic target or a clinical biomarker at present[1][3][5][6].
