LAMP5 antisense RNA 1 (LAMP5-AS1) is a long noncoding RNA transcribed from the antisense strand of the LAMP5 gene on chromosome 20. It is highly expressed in MLL-rearranged (mixed-lineage leukemia) cells and directly interacts with the lysine methyltransferase DOT1L to promote global H3K79 methylation, upregulating genes that maintain leukemia stemness and block differentiation. LAMP5-AS1 also regulates expression of the LAMP5 protein by recruiting the DOT1L complex to the LAMP5 locus, thereby inhibiting autophagic degradation of oncogenic MLL fusion proteins. High LAMP5-AS1 expression is a diagnostic and prognostic biomarker in MLL-rearranged leukemia. Its function is primarily epigenetic regulation in the context of oncogenesis, and it is under investigation as a potential therapeutic target in hematological malignancies. No direct interacting drugs are currently known, but its inhibition in cell and animal models impairs leukemia progression and promotes differentiation.
LAMP5-AS1 binds to and modulates the methyltransferase activity of DOT1L, enhancing global H3K79 methylation and facilitating the expression of genes critical to leukemia cell stemness. LAMP5-AS1 activates LAMP5 expression through direct interaction with DOT1L at the LAMP5 locus, which in turn inhibits autophagic degradation of oncogenic MLL fusion proteins.
