Overview

Langerhans cells (LCs) are specialized tissue-resident immune cells primarily found in the skin and mucosa. They function as antigen-presenting cells, capturing and processing antigens to initiate adaptive immune responses. They are also involved in immune regulation and maintaining tissue homeostasis. LCs express CD1a, langerin (CD207), and S100 protein and contain Birbeck granules. A key disease association is Langerhans Cell Histiocytosis (LCH), characterized by abnormal proliferation of LC-like cells, often driven by BRAF or MAP2K mutations.

Mechanism of action

Vemurafenib and dabrafenib inhibit BRAF V600E mutated kinases. Trametinib and cobimetinib inhibit MAP2K mutated kinases.
