Overview

Langerhans cells (LCs) are specialized, dendritic antigen-presenting cells located in the epidermis that serve as the primary immune sentinels of the skin (StatPearls, 2023). They are characterized by the expression of CD1a and Langerin (CD207) and are responsible for capturing allergens and transporting them to local lymph nodes (Nature Reviews Immunology, 2019). Once in the lymph nodes, LCs present these antigens to naive T-lymphocytes, leading to the generation of allergen-specific T-cells that orchestrate allergic inflammation or, under specific conditions, immune tolerance (Journal of Allergy and Clinical Immunology, 2020). This cellular axis is a critical component in the pathogenesis of atopic dermatitis and allergic contact dermatitis, where Th2-polarized T-cells drive the disease state (PubMed, 2021). Therapeutic strategies, such as epicutaneous immunotherapy (EPIT), specifically leverage the unique migratory and signaling properties of LCs to deliver allergens and promote the development of regulatory T-cells (Tregs) (DBV Technologies, 2024). By targeting this interaction, drugs aim to desensitize the immune system and reduce the hypersensitivity associated with specific allergens.
