Lanthionine synthetase C-like 2 (LANCL2) is a protein that acts as a receptor for abscisic acid (ABA), an endogenous mammalian hormone. Upon ligand binding, LANCL2 activates downstream signaling pathways involved in glycemic control and immune modulation. It is a promising therapeutic target for metabolic and inflammatory/autoimmune diseases. Activation of the LANCL2 pathway orchestrates immunoregulatory mechanisms via metabolic sensors such as AMPK, leading to increased fatty acid oxidation, mitochondrial metabolism, IL-2/CD25 signaling, Foxp3 expression, Treg induction/stability, and suppression of effector T-cell responses. Omilancor is currently undergoing late-stage clinical development for ulcerative colitis/Crohn’s disease.
