Overview

Large 60S subunit nuclear export GTPase 1 (LSG1) is an essential GTPase enzyme involved in the biogenesis and nuclear export of the 60S ribosomal subunit in eukaryotic cells[1][2]. LSG1 plays a crucial role in releasing NMD3 from the 60S ribosomal subunit after export from the nucleus to the cytoplasm, a step required for the proper assembly of functional ribosomes[2]. The protein is predominantly localized to the endoplasmic reticulum and, in some cases, found in discrete nuclear structures such as Cajal bodies[1]. LSG1 is part of the evolutionarily conserved YRG (YlqF Related GTPases) family, and its GTPase activity is essential for cell viability, nucleocytoplasmic transport, and rRNA maturation[1][3]. Direct drug targeting of LSG1 has not been reported, and it is not currently considered a therapeutic target in clinical practice.
