Large ribosomal subunit protein L4 (RPL4) is a key structural element of the 60S ribosomal subunit, encoded by the RPL4 gene on chromosome 15. It is required for ribosome assembly, interacts with ribosomal RNA, and lines the peptide exit tunnel in the ribosome. Its primary biological role is as a structural and functional constituent essential for translation in eukaryotic cells. RPL4 can undergo interactions with ribosome assembly chaperones (e.g., Acl4) to protect it from cellular degradation during biogenesis, and its proper assembly is crucial for cellular growth. Mutations or disruptions may lead to defects in ribosome biogenesis and are associated with disease states, notably some forms of anemia, cancer, and developmental disorders.
Not applicable; no drugs directly target human RPL4. In bacteria, ribosomal protein L4 is part of the macrolide binding site (not true of human RPL4).
