MRPL53 encodes a protein that is a component of the large (39S) subunit of the mitochondrial ribosome in mammals. It is encoded by nuclear DNA and imported into mitochondria, where it participates in translating mitochondrial mRNA to synthesize core subunits of the electron transport chain. Unlike cytoplasmic ribosomes, the mitoribosome has a higher protein-to-rRNA ratio and lacks a 5S rRNA. MRPL53 is expressed across numerous tissues and is required for proper mitochondrial translation and energy production. Impairment of mitochondrial ribosomal subunits, including MRPL53, can result in decreased energy production and mitochondrial disease. However, MRPL53 itself is not recognized as a therapeutic target or used as a clinical biomarker.
