Overview

Large ribosomal subunit protein uL3 (uL3, also known as RPL3 in humans) is a universally conserved component of the large ribosomal subunit. It plays a critical structural and functional role within the ribosome, responsible for protein synthesis. uL3 is essential for maintaining the architecture of the peptidyl transferase center (PTC), which catalyzes peptide bond formation. It binds directly to rRNA and helps organize the catalytic core of the ribosome. Mutations in uL3 can disrupt PTC structure and function. It participates broadly across cellular biology including ribosomal assembly, mRNA decay, and gene expression. A paralogue, RPL3L/uL3-like, exists that is specifically expressed in heart and skeletal muscle.
