Large tumor suppressor kinase 1 (LATS1) is a serine/threonine kinase and core component of the Hippo signaling pathway, critical for regulating cell proliferation, apoptosis, and organ size. LATS1 phosphorylates and inactivates YAP and TAZ, preventing their nuclear localization and transcriptional activation of proliferative genes. Mutations or loss of LATS1 function are associated with increased susceptibility to various cancers due to loss of cell cycle regulation and failure of apoptosis, supported by mouse knockout studies that show increased tumorigenesis. LATS1 also interacts with cytoskeletal proteins, directly binds and inhibits actin polymerization, and localizes to structures such as centrosomes and mitotic apparatus. As a negative regulator of cell migration, LATS1's suppression of metastasis further cements its tumor suppressor role[1][2][3][4][5].
