Overview

Late cornified envelope-like proline-rich protein 1 (LELP1) is a small, proline-rich protein encoded by the LELP1 gene on chromosome 1q21, belonging to the cornifin (SPRR) protein family[1][3][9]. LELP1 is primarily expressed in the skin, where it is thought to function in the formation and structural integrity of the epidermal cornified envelope, contributing to barrier function and epidermal differentiation[2][3][9]. Increased expression of LELP1 has been observed in atopic dermatitis skin, with evidence suggesting that variants in the LELP1 gene (e.g., rs7534334 SNP) may influence serum IgE levels and disease course in atopic dermatitis and atopic asthma[2][11]. Its precise molecular function remains unknown, but it is not classified as a receptor, enzyme, transporter, or major therapeutic target[2][3][1][8]. LELP1 is a potential biomarker for skin barrier function and disease in dermatological research, but no drugs target LELP1 directly.
