Overview

Late cornified envelope protein 1C (LCE1C) is a human protein encoded by the LCE1C gene, part of the late cornified envelope cluster on chromosome 1q21[3][7]. It serves as a precursor to the cornified envelope of the stratum corneum, contributing to the barrier function of the epidermis[2][3][7]. The protein is implicated in keratinization—a process vital for skin formation and integrity[3]. LCE1C is a transcriptional target downstream of p63, particularly regulated by phosphorylation of TAp63, and it plays a role in epithelial differentiation, cell cycle regulation, and acts as a tumor suppressor by interacting with and translocating PRMT5 from the nucleus to the cytoplasm, reducing PRMT5 nuclear function[1][5][6]. Diseases associated with variants in LCE1C include psoriasis and certain cataracts[3]. There are currently no drugs or therapies directly known to target LCE1C.
