Late cornified envelope protein 1D (LCE1D) is a member of the late cornified envelope (LCE) protein family within the epidermal differentiation complex (EDC) on human chromosome 1q21[2][1]. It is a structural protein component of the cornified envelope—a highly cross-linked protein and lipid structure that provides epidermal barrier functions, forming during the terminal differentiation of keratinocytes in the skin[1][11][2]. LCE1D enables identical protein binding activity, is involved in cognition, and acts upstream of or within the cellular response to calcium ions[1][5][9]. It has not been described as a traditional therapeutic target (such as a receptor, enzyme, or transporter). The closest known clinical association is to rare inherited skin conditions such as focal nonepidermolytic palmoplantar keratoderma[1]. No approved drugs or targeted therapies are known to interact with this protein, and it is not currently recognized as a biomarker or as having important safety concerns in the clinical context.
