Overview

Late cornified envelope protein 2B (LCE2B) is a structural protein that acts as a precursor for the cornified envelope, which forms the outermost barrier of the skin (stratum corneum)[1][4]. It belongs to the late cornified envelope (LCE) protein family, a component of the epidermal differentiation complex (EDC) on chromosome 1q21[3][7]. LCE2B is highly expressed in the skin, especially during terminal keratinocyte differentiation, and is upregulated in certain skin conditions such as psoriasis[1][11]. The protein helps establish and reinforce the skin’s barrier function, crucial for protecting underlying tissues and maintaining hydration. While its primary function is structural, genetic variation or altered expression of LCE2B and related proteins has been implicated in several skin diseases, notably psoriasis[1][3]. No drugs are currently known to specifically target LCE2B, and it is not considered a classical therapeutic target such as a receptor, enzyme, ion channel, or transporter[1][3][11].
