Late cornified envelope protein 2D (LCE2D) is a member of the late cornified envelope (LCE) protein family, encoded within the epidermal differentiation complex (EDC) on chromosome 1[1][4][5]. This protein functions as a structural precursor in the formation of the cornified envelope, the protein-lipid barrier that forms the outermost layer of the skin (stratum corneum), and is involved in keratinocyte terminal differentiation and cornification[1][3][4]. Expression of LCE2D is largely restricted to the skin, particularly differentiated keratinocytes, and is regulated by lineage-specific transcription factors involved in epidermal biology[2]. LCE2D is not considered a conventional therapeutic target (such as a receptor, enzyme, or transporter), nor are there characterized small-molecule drugs or biologics known to directly interact with it. Mutations or dysregulation of LCE family genes, including LCE2D, have been studied in the context of skin barrier function and certain dermatological diseases[2][4].
