Late cornified envelope protein 6A (LCE6A) is a member of the LCE protein family localized within the epidermal differentiation complex on human chromosome 1q21. LCE6A is involved in the late stages of cornified envelope formation in differentiated keratinocytes, contributing to the mechanical integrity and barrier function of the skin’s outer layer. Its expression is induced in response to calcium signaling and ultraviolet irradiation during terminal differentiation. LCE family members—including LCE6A—are incorporated into the cornified envelope by transglutaminase-mediated cross-linking, and participate in the modulation of the epidermal barrier’s quality and resistance to environmental insult. There are currently no documented disease mutations, drug interactions, nor established use as a biomarker or therapeutic target for LCE6A in clinical practice.
