Late cornified envelope pseudogene 2 (LCEP2) is part of a group of pseudogenes located within the epidermal differentiation complex (EDC), which also contains numerous genes encoding late envelope proteins (LEPs). These functional LEPs are expressed very late during epidermal differentiation and are important for the final stages of cornified envelope maturation, contributing to the skin’s barrier properties by being incorporated via epidermal transglutaminase-mediated cross-linking[1][3]. However, LCEP2 itself is not thought to encode a functional protein; as a pseudogene, it is a noncoding remnant that does not contribute directly to protein synthesis or known biological functions. No evidence supports LCEP2 as a therapeutic target, biomarker, or disease driver, and it does not interact with drugs or have a defined mechanism of action. The presence of the LCEP2 pseudogene reflects the evolutionary history and gene duplication events in the EDC region[3].
