Latrophilin Receptor LAT-1/LAT-2 (LPHN1/LAT-1 & LPHN2/LAT-2)

Target
LPHN1/LAT-1 & LPHN2/LAT-2
Molecular classification
Adhesion G-protein-coupled receptor (aGPCR), GPCR, Receptor
01

Overview

Latrophilins (LAT-1 and LAT-2) are adhesion G-protein-coupled receptors (GPCRs) crucial for cell signaling, synapse formation, and tissue polarity. LAT-1 is primarily brain-specific and involved in synapse maturation and exocytosis. LAT-2 is ubiquitously expressed and implicated in breast cancer biology and postsynaptic target recognition. Both interact with ligands like alpha-latrotoxin and teneurins, activating G-protein pathways and influencing calcium release. In C. elegans, LAT-1 is vital for establishing anterior-posterior tissue polarity.

Mechanism of action

Ligand binding induces receptor reassociation, activating intracellular G-protein–mediated pathways and releasing Ca²⁺ from internal stores, leading to neurotransmitter or hormone secretion/exocytosis.
