Overview

LATS2 antisense RNA 1 (LATS2-AS1) is a long non-coding RNA (lncRNA) transcribed antisense to the LATS2 gene locus. It is not a receptor, enzyme, or classical drug target, but exerts regulatory influence on its neighboring gene LATS2, typically acting as a tumor suppressor in gastric cancer. LATS2-AS1 appears to upregulate LATS2 expression and thereby suppresses YAP1/TAZ oncogenic signaling in the Hippo pathway, leading to decreased cancer cell proliferation, induction of G0/G1 cell cycle arrest, and reduced cell invasion and migration. Its expression is significantly reduced in gastric cancer tissues; its levels are correlated with disease progression, suggesting potential value as a diagnostic biomarker and a putative future therapeutic target in oncology.

Mechanism of action

Not applicable for drugs, as no direct modulators of LATS2-AS1 are in clinical use. Mechanistically, LATS2-AS1 modulates LATS2 gene expression and enhances LATS2-mediated Hippo/YAP pathway suppression by binding to EZH2 (Enhancer of zeste homolog 2) as a regulatory lncRNA.
