Overview

Layilin is a type I transmembrane protein and a cell surface receptor with a domain homologous to C-type lectin carbohydrate-recognition regions[1][2]. It serves as a receptor for hyaluronan, a major extracellular matrix glycosaminoglycan, playing a role in cell–extracellular matrix adhesion, cell migration, actin cytoskeleton reorganization, and immune cell function[1][2]. Layilin interacts with talin and possibly other ERM family proteins, linking the extracellular environment to the cytoskeleton and participating in dynamic membrane ruffling events and immune modulation. Expression of LAYN has been correlated with prognosis and immune infiltration in different cancers, especially colorectal and gastric cancer, and may be important in the regulation of tumor-associated macrophages, dendritic cells, and T cell exhaustion[3].
