LBX1 antisense RNA 1 (LBX1-AS1)

Target
LBX1-AS1
Molecular classification
Long non-coding RNA (lncRNA), Antisense RNA
01

Overview

LBX1 antisense RNA 1 (LBX1-AS1) is a long non-coding RNA (lncRNA) transcribed in proximity to the LBX1 locus, head-to-head with the LBX1 gene. LBX1-AS1 regulates gene expression by sponging microRNAs such as miR-182-5p, leading to upregulation of tumor suppressor genes like FOXO3 and inhibition of cell proliferation and invasion in oral squamous cell carcinoma. It also impacts epigenetic regulation via altered CTCF binding, affecting DNA methylation and the expression of neighboring genes. LBX1-AS1 is associated with disease processes including cancer and glaucoma, serving as a potential biomarker for disease progression and prognosis. LBX1-AS1 is not considered a classical druggable target but is important in regulatory RNA biology and disease association studies.

02

Mechanism of action

Acts as a competitive endogenous RNA (ceRNA) inhibiting miR-182-5p and upregulating FOXO3, a tumor suppressor gene. May affect gene expression through modulation of CTCF binding and DNA methylation.

03

Biological functions

Gene expression regulationTumor suppression (demonstrated in oral squamous cell carcinoma)miRNA sponging (miR-182-5p)
04

Disease associations

Cancer (oral squamous cell carcinoma)Primary open-angle glaucoma (association with gene polymorphisms)Potential regulatory roles in other conditions linked to LBX1, e.g., adolescent idiopathic scoliosis, but LBX1-AS1’s specific contribution is less established
05

Safety considerations

No direct safety concerns are described, as LBX1-AS1 is not a direct target of any approved therapiesTherapeutic modulation of noncoding RNAs may have off-target regulatory effects
06

Biomarkers

LBX1-AS1 expression can serve as a biomarker for tumor suppression activity and prognosis in oral squamous cell carcinomaPotentially a biomarker for association studies in glaucoma
