Overview

It acts as a negative regulator of NF-κB transcriptional activity, functioning as a tumor suppressor by inhibiting factors involved in inflammation, cell survival, and proliferation[2][4]. LDOC1 is commonly down-regulated in multiple cancer types, often due to epigenetic silencing via promoter methylation. Restoring LDOC1 activity can suppress NF-κB-dependent cytokine production (e.g., IL-6) and may resensitize tumor cells to chemotherapy[2]. LDOC1 expression is associated with clinical outcomes in certain cancers, making it a candidate prognostic biomarker. Direct pharmacological targeting is not currently available, but its regulation via epigenetic therapies is under investigation[2].
