Overview

Left-right determination factor 1 (LEFTY1) is a secreted protein and ligand of the transforming growth factor-beta (TGF-beta) superfamily that is essential for the proper establishment of left-right asymmetry of visceral organs during embryonic development. LEFTY1 functions primarily by acting as an antagonist to the Nodal signaling pathway, thereby restricting the actions of Nodal and related factors (such as LEFTY2) to specific regions and ensuring correct organ patterning. It is produced as a preproprotein and secreted after proteolytic cleavage. Mutations or dysregulation of LEFTY1 can lead to severe congenital abnormalities, such as situs inversus or left pulmonary isomerism, due to failure in establishing appropriate left-right cues, particularly affecting the heart and lungs. LEFTY1 is closely related to its paralog LEFTY2 and is not considered a common direct therapeutic target, nor are there drugs known to specifically target it[1][2][3][7].
