Overview

Legumain pseudogene 1 (LGMNP1) is a pseudogene located on chromosome 13, related to the functional legumain gene (LGMN) that encodes a cysteine protease involved in diverse biological processes, including cancer progression[1][4]. Unlike the protein-coding gene, LGMNP1 does not code for a functional protein but acts as a non-coding RNA. LGMNP1 is upregulated in glioblastoma cells following exposure to ionizing radiation, and its overexpression enhances cellular resistance to radiotherapy by increasing DNA repair capacity and reducing apoptosis. Evidence suggests LGMNP1 may exert its effects by influencing the expression or function of its parental gene, legumain, through regulatory mechanisms typical of pseudogenes[1][4]. While LGMNP1 is not itself a direct therapeutic target like a receptor, enzyme, or transporter, it may serve as a novel indirect target to sensitize glioblastoma to radiotherapy. No drugs currently target LGMNP1, and there are no known direct safety concerns associated with it; its primary relevance is as a potential biomarker or target for overcoming treatment resistance in cancer[1][4].
