Overview

The amastigote form of Leishmania amazonensis is a small, spherical to ovoid, intracellular stage of the parasite that replicates within macrophages in mammalian hosts[7][1]. Amastigotes possess a large nucleus, kinetoplast, and a very short, usually non-motile flagellum[2][7]. This stage is highly adapted to survive the hostile environment within host phagolysosomes, exhibiting resistance to oxidative and nitrosative stress, partly through upregulation of antioxidant enzymes such as tryparedoxin peroxidase[3][5]. The amastigote form is responsible for the clinical manifestations of leishmaniasis, particularly cutaneous and diffuse cutaneous forms, and is the target of all current antiparasitic therapies. Its persistence in macrophages contributes to chronic infection and represents a major challenge for treatment, as the parasite can evade many microbicidal mechanisms of host cells and resist pharmacologic eradication[3][5][6].
