Overview

Leishmania major oligopeptidase B (LmOpB) is a key cytosolic serine protease belonging to the S9 family, essential for the pathogenesis and survival of the Leishmania parasite within host macrophages. Unlike many other members of its family that exhibit prolyl endopeptidase activity, LmOpB specifically cleaves peptide bonds following basic amino acid residues such as arginine and lysine. It functions as a significant virulence factor by modulating the host's immune response and degrading host signaling molecules that would otherwise trigger parasite clearance. The enzyme is involved in the regulation of parasite differentiation and is crucial for maintaining the infection cycle in the vertebrate host. Because its substrate specificity differs from that of human prolyl oligopeptidases, LmOpB is considered a promising target for the development of selective antileishmanial drugs. Inhibiting this enzyme aims to reduce the parasite's ability to evade the host immune system and limit the progression of cutaneous leishmaniasis.
