Overview

The Leishmania major peptide-Major Histocompatibility Complex (MHC) class I complex is a molecular assembly found on the surface of antigen-presenting cells, such as dendritic cells and macrophages, during an infection with the protozoan parasite Leishmania major. This complex consists of a short peptide fragment derived from parasite proteins—such as LACK (Leishmania homolog of receptors for activated C kinase), KMP-11 (Kinetoplastid membrane protein-11), or TSA (Thiol-specific antioxidant)—bound within the groove of an MHC class I molecule (PMID: 11160251). Its primary biological function is to present these foreign antigens to the T-cell receptors (TCRs) of CD8+ T lymphocytes, thereby triggering a cellular immune response characterized by the production of interferon-gamma (IFN-gamma) and the destruction of infected cells (PMID: 15661044). In the context of cutaneous leishmaniasis, the efficient presentation of these complexes is essential for parasite clearance and the establishment of long-term immunity. Consequently, these complexes are major targets for the development of subunit vaccines and immunotherapies aimed at enhancing the host's ability to recognize and eliminate the parasite (PMID: 10590271).
