Overview

Leishmania-specific antigens are a heterogeneous group of proteins and glycoconjugates expressed by various species of the Leishmania genus, which are the causative agents of leishmaniasis. These antigens, including GP63 (a zinc-dependent metalloprotease), K39 (a kinesin-related protein), and LACK (Leishmania homolog of receptors for activated C kinase), are critical for the parasite's ability to infect host macrophages and evade the immune system (Yao et al., 2003, PMID: 12692232). In the field of vaccinology, these antigens are targeted to induce a protective Th1-type immune response, which is essential for controlling the intracellular infection (Coler & Reed, 2005, PMID: 15916607). Additionally, specific antigens like rK39 are widely used in rapid diagnostic tests to detect visceral leishmaniasis due to their high sensitivity and specificity for anti-Leishmania antibodies (Burns et al., 1993, PMID: 8419301). While several antigen-based vaccines have reached clinical trials, challenges such as the requirement for potent adjuvants and the need for long-lasting cellular memory remain (Gillespie et al., 2016, PMID: 27273243).
