Overview

LEMD1 (LEM domain-containing protein 1) is a nuclear envelope-associated protein characterized by the presence of a LEM domain, which mediates interactions with nuclear lamins, chromatin, and various proteins involved in nuclear structure and function. LEMD1 is predominantly localized at the inner nuclear membrane and is involved in the assembly and disassembly of the nuclear envelope during cell division, maintenance of nuclear integrity, and organization of chromatin. High expression of LEMD1 is observed in several cancers (including triple-negative breast cancer and thyroid cancer), where it promotes proliferation, migration, and epithelial-mesenchymal transition through activation of ERK and Wnt/β-catenin pathways. As a member of the cancer/testis antigen family, LEMD1 is also under investigation as a diagnostic and potential therapeutic biomarker in oncology. No known drugs directly target LEMD1 as of the current literature.
