Lens epithelium-derived growth factor (LEDGF), also known as PSIP1 or LEDGF/p75, is a chromatin-binding protein and transcriptional coactivator. It plays critical roles in cellular survival, gene regulation, and disease processes such as HIV infection and leukemia. Targeting the interaction between LEDGF/p75’s IBD and HIV integrase has been explored therapeutically because disrupting this interface inhibits viral replication. Aberrant expression or function of LEDGF can contribute to oncogenesis—notably mixed-lineage leukemia.
