Lepidoglyphus destructor allergen refers to a group of proteins produced by the storage mite Lepidoglyphus destructor, a common inhabitant of stored grain, hay, and damp household environments. These allergens, particularly the major allergen Lep d 2, are significant triggers of IgE-mediated hypersensitivity reactions, including allergic rhinitis, conjunctivitis, and bronchial asthma (PMID: 11030345). Lep d 2 belongs to the Niemann-Pick type C2 (NPC2) protein family and functions as a lipid-binding protein, a characteristic that may contribute to its high allergenicity (UniProt P81528). In clinical practice, these allergens are the primary targets for allergen-specific immunotherapy (AIT), where they are used to induce immunological tolerance in sensitized individuals. The therapeutic mechanism involves shifting the immune response from a Th2-mediated allergic state to a Th1 and regulatory T-cell (Treg) dominated profile, accompanied by the production of blocking IgG4 antibodies (PMID: 30033913). While AIT is effective for long-term symptom relief, the administration of Lepidoglyphus destructor extracts carries a risk of systemic allergic reactions, including anaphylaxis, necessitating careful dose titration and medical supervision (World Allergy Organization).
