The leptin receptor is a transmembrane receptor that mediates the effects of leptin, a hormone primarily involved in regulating metabolism, appetite, and energy balance. It is most highly expressed on hypothalamic neurons and plays roles in bone metabolism, immunity, and reproductive function. Alternative splicing results in multiple isoforms with varying signaling capabilities.
Agonism of the leptin receptor leads to activation of downstream JAK/STAT pathways, resulting in transcriptional changes that suppress appetite and increase energy expenditure. Antagonism is being investigated for other potential benefits.
