Leptospira interrogans serogroup Icterohaemorrhagiae antigens represent the collection of immunogenic molecules, primarily surface-exposed proteins and lipopolysaccharides, found on the pathogenic spirochete Leptospira interrogans within the Icterohaemorrhagiae serogroup (Picardeau, 2017). These antigens, such as the highly conserved lipoprotein LipL32 and the immunoglobulin-like proteins LigA and LigB, are fundamental to the bacterium's virulence, facilitating attachment to host extracellular matrix components and enabling immune evasion (Haake & Levett, 2015). They serve as the primary targets for the host's immune response and are the basis for diagnostic serology and vaccine development (Adler & de la Peña Moctezuma, 2010). In the context of therapeutics, these antigens are targeted by inactivated whole-cell vaccines and experimental recombinant vaccines designed to prevent leptospirosis and its severe form, Weil's disease (Ko et al., 2009). While antibiotics like penicillin and doxycycline are used to clear the infection by targeting bacterial protein synthesis or cell wall integrity, the antigens themselves are the focus of prophylactic strategies aimed at inducing long-term immunity. The Icterohaemorrhagiae serogroup is particularly significant as it is frequently associated with the most severe clinical manifestations of the disease in humans. Effective targeting of these antigens is complicated by the high degree of antigenic diversity among different Leptospira serovars, often necessitating serogroup-specific vaccine formulations.
Vaccines containing these antigens stimulate the production of specific antibodies and activate cellular immune responses to prevent bacterial colonization and dissemination.
