Overview

Leucine-rich repeat and calponin homology domain-containing protein 2 (LRCH2) is a cytoskeletal regulator protein characterized by multiple N-terminal leucine-rich repeats (LRR) and a C-terminal calponin homology (CH) domain, conserved across animal species. The LRR domain mediates protein-protein interactions, while the CH domain structurally associates with actin filaments. LRCH2 acts as a scaffold protein implicated in cytoskeletal organization, cell division, and migration, particularly in leukocytes, suggesting roles in immune surveillance and actin cytoskeleton dynamics. A similar protein in Drosophila shows related cytoskeletal functions. LRCH2 is not currently recognized as a druggable therapeutic target, and alternative splicing produces several transcript variants. Diseases associated with LRCH2 include demyelinating polyneuropathy and spinocerebellar ataxia 44, though the pathophysiological mechanisms are not well understood.
