Leucine-rich repeat and fibronectin type III domain-containing protein 1 (LRFN1, also known as SALM2) is a neuronal transmembrane glycoprotein of the SALM/Lrfn protein family predominantly expressed in the central nervous system. It features extracellular leucine-rich repeats, immunoglobulin-like and fibronectin type III motifs, and a cytoplasmic PDZ-binding sequence. LRFN1 interacts with postsynaptic scaffolding proteins such as PSD-95, and it is involved in the organization and maturation of excitatory synapses, regulation of postsynaptic density assembly, and neurite outgrowth. While not a classic therapeutic target (receptor, transporter, enzyme), LRFN1 plays roles in neuronal function and has been linked to neurodevelopmental disorders, epilepsy, and inclusion in gene expression panels for cancer prognosis, but no targeting drugs or therapeutic mechanism of action is currently reported[2][4][6][8].
