Leucine-rich repeat and guanylate kinase domain-containing protein (LRGUK) is a cytosolic protein characterized by the presence of leucine-rich repeats and a guanylate kinase-like domain[1][2][4][3]. It is highly expressed in testis, especially in germ cells undergoing spermatogenesis, where it is essential for proper sperm assembly, encompassing roles in acrosome attachment, manchette function, sperm head shaping, and axoneme development[2][4][3]. Mutations in LRGUK disrupt these processes, leading to abnormal sperm morphology and male infertility, as shown in animal models[2]. While predicted to possess GMP kinase activity, it is not classified as an enzyme, receptor, transporter, or canonical therapeutic target family[3][4]. No currently known drugs target LRGUK, nor is it validated as a clinical biomarker or implicated in therapy; its main significance is as an essential structural and assembly protein for male germ cell function and fertility[2][3][4].
