Overview

LINGO-1 (Leucine-rich repeat and Ig domain-containing Nogo receptor-interacting protein 1) is a transmembrane protein predominantly expressed in the central nervous system (CNS). It acts as a negative regulator of myelination and inhibits axonal regeneration. LINGO-1 interacts with Nogo receptor complexes and myelin-associated inhibitors to activate RhoA-GTP signaling, leading to inhibition of neurite outgrowth and oligodendrocyte differentiation. It also modulates BK channel activity. Elevated LINGO-1 levels are implicated in neurodegenerative disorders such as Multiple Sclerosis, Parkinson's Disease, Essential Tremor and Alzheimer's Disease, making it a potential therapeutic target for promoting nerve repair and remyelination.

Mechanism of action

Pharmacological inhibition or antibody-mediated blockade against LINGO-1 promote nerve repair/remyelination after CNS injury by blocking its inhibitory effects on oligodendrocyte maturation and axonal regeneration. Anti-LINGO-1 antibodies bind to LINGO-1, preventing its interaction with NgR1 and downstream signaling pathways.
