Overview

Leucine-rich repeat and immunoglobulin domain-containing protein 1, commonly known as LINGO-1, is a central nervous system-specific transmembrane protein that functions as a potent negative regulator of axonal regeneration and oligodendrocyte differentiation. It acts as a crucial component of the Nogo-66 receptor complex (NgR1), partnering with co-receptors such as p75 or TROY to activate the RhoA signaling pathway, which inhibits myelin formation and neurite outgrowth. This protein is expressed exclusively in the brain and spinal cord, predominantly in neurons and oligodendrocytes. LINGO-1 is a significant therapeutic target for neurodegenerative and demyelinating diseases like Multiple Sclerosis, Parkinson's disease, and Essential tremor, where its overactivity or presence prevents natural repair mechanisms. Therapeutic strategies have focused on LINGO-1 antagonists, most notably the monoclonal antibody opicinumab, to promote remyelination and neuroprotection. Despite robust results in animal models showing enhanced nerve repair, human clinical trials have encountered significant challenges in meeting primary efficacy endpoints, highlighting the complexity of its role in human neurobiology.
