Overview

LINGO-1 is a transmembrane protein primarily expressed in the central nervous system, acting as a negative regulator of axonal regeneration, neurite outgrowth, and myelination. It functions within the Nogo receptor signaling complex, inhibiting neurite extension and myelin formation through RhoA/ROCK pathway activation. LINGO-1 is implicated in neurodegenerative diseases like multiple sclerosis, Parkinson's disease, and Alzheimer's disease. It modulates BK channel activity and plays a role in amyloid-beta precursor protein processing.

Mechanism of action

Inhibition of LINGO-1 enhances survival/functionality of dopaminergic neurons and promotes remyelination after demyelinating injury/disease states. Acts as a functional component of the Nogo receptor signaling complex (NgR/p75 or NgR/TROY complexes). Inhibits axonal regeneration via RhoA activation pathway when part of the ternary complex with NgR1 and p75/TROY receptors. This leads to downstream activation of Rho-associated kinase (ROCK), blocking neurite extension and myelin formation
