Leucine-rich repeat and IQ motif-containing protein 3 (LRRIQ3) is a protein-coding gene that encodes a membrane-associated protein featuring three leucine-rich repeats and a single IQ calmodulin-binding motif. The protein is expressed at low to moderate levels in human tissues, with highest expression in the testes, some immune cells, kidney, glands, and epididymis. LRRIQ3 is involved in protein-protein interactions with molecules such as LYN, NCK2, GNB4, and ABL1, all of which are linked to signaling and cytoskeleton regulation. Genome-wide association studies implicate genetic variation in LRRIQ3 in susceptibility to chronic pain and major depressive disorder. Its transcript levels have been proposed as part of a predictive gene signature for rectal cancer response to chemoradiotherapy, and its downregulation has been observed in several cancer types, supporting its role as a novel biomarker for both neuropsychiatric and oncological contexts. No validated drugs or direct therapeutic targeting information is currently available.
